Transverse Sectioning of Mature Rice (<em>Oryza sativa</em> L.) Kernels for Scanning Electron Microscopy Imaging Using Pipette Tips as Immobilization Support

Starch granules (SGs) exhibit different morphologies depending on the plant species, especially in endosperm of Poaceae family. Endosperm phenotyping can be used to classify genotypes based SG morphotype using scanning electron microscopic (SEM) analysis. SGs visualized SEM by slicing through kernel (pericarp, aleurone layers, and endosperm) exposing organellar contents. Current methods require rice embedded plastic resin sectioned a microtome or truncated pipette tip hand razor blade. The former method requires specialized equipment is time-consuming, while latter introduces new host problems genotype. Chalky varieties, particularly, pose problem for this type sectioning due friable nature their tissue.
